require_once()

would have caused a fatal error if the file was not found !!

so ... maybe a PHP bug?
what is the PHP version?

maybe you could open file:

JPATH_SITE.DS.'components'.DS.'com_flexicontent'.DS.'helpers'.DS.'route.php'

and temporarily place an die('reached'); (or maybe something less radical like writting something in a file and then refreshing the page that gives the erro 500
